Snalysis of Sterigmatocystin by Enzyme-Linked Immunosorbent Assay

Abstract

|
To establish method which is simpler, more rapid, more sensitive, and more specific than thin-layer chromatograpy (TLC) or high performance liquid chromatography (HPLC) which analyze sterigmatocystin, we establish ELISA after producing of high
titer of
antibody against sterigmatocystin and then obtained following results from anaysis of sterigmatocystin produced by Aspergillus versicolor ATCC 18643.
1) Two of three rabbits which had been immunized with sterigmatocystin-hemiacetal but didn't cross react with other sterigmatocystin analogues.
2) The antibodies were specific for sterigmatocystiong and sterigmatocystin-hemiacetal but didn't cross react with other sterigmatocystin analogues.
3) It took only 2 hours to perform the ELISA procedure.
4) SLS media were more productive of sterigmatocystin than YES media and the production of sterigmatocystin was maximal between 9th day and 12th day.
5) Sterigmatocystin was more produced in mycelia (254 ㎍/ml).
